Requirements include at least four fluorescence channels, a high-sensitivity confocal detector, confocal and wide-field modes, a cryo-compatible air objective with at least 50× magnification, and a motorised cryo-stage that handles vitrified samples below −150 °C without warming or significant moisture contamination.
